The Escherichia coli araBAD operon consists of three genes encoding three enzymes that convert L-arabinose to D-xylulose-5 phosphate. In this paper we report that the genes of the E. coli araBAD operon have been expressed in Saccharomyces cerevisiae using strong promoters from genes encoding S. cerevisiae glycolytic enzymes (pyruvate kinase, phosphoglucose isomerase, and phosphoglycerol kinase). The expression of these cloned genes in yeast was demonstrated by the presence of the active enzymes encoded by these cloned genes and by the presence of the corresponding mRNAs in the new host. The level of expression of L-ribulokinase (araB) and L-ribulose-5-phosphate 4-epimerase (araD) in S. cerevisiae was relatively high, with greater than 70% of the activity of the enzymes in wild type E. coli. On the other hand, the expression of L-arabinose isomerase (araA) reached only 10% of the activity of the same enzyme in wild type E. coli. Nevertheless, S. cerevisiae, bearing the cloned L-arabinose isomerase gene, converted L-arabinose to detectable levels of L-ribulose during fermentation. However, S. cerevisiae bearing all three genes (araA, araB, and araD) was not able to produce detectable amount of ethanol from L-arabinose. We speculate that factors such as pH, temperature, and competitive inhibition could reduce the activity of these enzymes to a lower level during fermentation compared to their activity measured in vitro. Thus, the ethanol produced from L-arabinose by recombinant yeast containing the expressed BAD genes is most likely totally consumed by the cell to maintain viability. © 2001 Elsevier Science Inc. Lactic acid bacteria display a relatively simple metabolism wherein the sugar is converted mainly to lactic acid. The extensive knowledge of metabolic pathways and the increasing information of the genes involved allows for the rerouting of natural metabolic pathways by genetic and physiological engineering. We discuss several examples of metabolic engineering of Lactococcus lactis for the prod...

During the mashing process of brewing, activity of the amylolytic enzymes decays due to the high temperatures used to gelatinise the starch. Because the different enzymes produce different sugars, high temperatures can be exploited to modify the fermentability of resulting worts. This is especially useful when producing low alcohol beers. The expression a.exp(b.t)-c.exp(d.t) (where t is the tem...

We have made experimental studies into the enzymatic hydrolysis of cellobiose within the temperature range of 40°C to 70°C at pH 4.9, by using b-1,4-glucosidase from Aspergillus niger. At 70°C there was significant enzyme deactivation, which could be fitted to a potential deactivation model with values of n equal to 1.09 and kd to 0.1564 (g/l) 20.09 min, whereas the rate of hydrolysis could be ...

In general the biodegradation of nonchlorinated aliphatic and aromatic hydrocarbons is influenced by their bioavailability. Hydrocarbons are very poorly soluble in water. They are easily adsorbed to clay or humus fractions in the soil, and pass very slowly to the aqueous phase, where they are metabolised by microorganisms. Surfactants that increase their solubility and improve their bioavailabi...

This paper deals with the sequential diagnosis approach and the associated test sequencing problem (i.e. the problem of generating the sequence of tests required to reach a diagnostic conclusion at minimum cost). The traditional test sequencing problem is generalized here to include asymmetrical and multi-valued tests. We present the sequential diagnosis tool (SDT) that enables the user to gene...
